Вентральная тегментальная область (VTA) оценивает значимость стимулов окружающей среды и обеспечивает дофаминергическую иннервацию во многих областях мозга, пораженных острым и хроническим воздействием этанола. Хотя в первую очередь это связано с поощрением и усилением стимулов, последние данные указывают на роль VTA в отвращении. Действия этанола в VTA могут вызвать нейроадаптацию, приводящую к снижению аверсивных реакций на алкоголь и относительному увеличению вознаграждающих реакций. В поисках эффективных фармакотерапий для лечения злоупотребления алкоголем и алкоголизма признание этого дисбаланса может выявить новые стратегии. В дополнение к традиционной фармакотерапии рецепторов/ионных каналов эпигенетические факторы, которые контролируют нейроадаптацию к лечению хроническим этанолом, могут быть нацелены на развитие терапевтических подходов к восстановлению баланса. Кроме того, при изучении методов лечения для устранения дисбаланса вознаграждения / отвращения в действии алкоголя в VTA необходимо учитывать половые различия, чтобы обеспечить эффективное лечение как для мужчин, так и для женщин. Эти принципы применимы к VTA-центричному подходу к терапии, но должны оставаться верными, когда речь идет об общем подходе к разработке нейроактивных препаратов для лечения расстройств, связанных с употреблением алкоголя. Хотя функции VTA сами по себе сложны, это полезная модельная система для оценки дисбаланса вознаграждения / отвращения, который возникает при воздействии этанола и может быть использован для обеспечения новых идей в усилиях по разработке новых лекарств для лечения алкоголизма.
